You need the third level of Alohomora, the lockpicking spell: return 22 Demiguise statues to Gladwin Moon in the side quest The Man Behind the Moons. The main quest The Polyjuice Plot takes you to the Headmaster's Office, which is shut before it.

Fast travel to the Trophy Room Floo Flame in the Grand Staircase.
Walk straight across the Trophy Room, turn right through the gate and climb the stairs to the Headmaster's Office.
Open the level 2 door in the office, climb outside to the next door and open its level 3 lock.
Go up into the upper study.
